Synthesis of Dexlansoprazole
Abstract
Dexlansoprazole,a chiral proton pump inhibitor, was synthesized from 2-mercaptobenzimidazole by condensation with 2-(chloromethyl)-3-methyl-4-(2,2,2-trifluoroethoxy)pyridine hydrochloride to give 2-{[3-methyl-4-(2,2,2-trifluoroethoxy)-2-pyridyl]methylthio}benzimidazole,which was subjected to vanadiumcatalyzed asymmetrical oxidation with cumene hydroperoxide. The overall yield was 35%.This synthetic method is suitable for industrial application for advantages such as moderate conditions, convenient operations, and high yield.
