Pharmacokinetics of Dexlansoprazole Capsules with a Biphase Release in Beagle Dogs
Abstract
A LC-MS/MS method was established for the determination of dexlansoprazole in dog plasma. The pharmacokinetics of dexlansoprazole delayed-release capsules(Dexilant) in Beagle dogs was investigated.AC_(18) column was used with the mobile phase of methanol-water(containing 10 mmol/L ammonium formate,70:30),and omeprazole was used as the internal standard. The calibration curve was linear in the range of 5-2 000 ng/ml.The extraction recoveries were 105.7%-123.1%.The intra-and inter-batch RSDs were less than 10%.After oral administration to Beagle dogs,a biphase-release model was observed. The main pharmacokinetic parameters were as follows:t_(1/2)(0.6±0.1) h, AUC_(0-∞)(2 019±176)ng·ml~(-1)·h,c_(max)(810.9±194.0)ng/ml.
