Preparation and release in vitro of colon-targeted tablet of indissolvable drug Lansoprazole
Abstract
OBJECTIVE To study the preparation of lansoprazole colon-targeted tablet(Lansoprazole colon-targeted,LPZ-CT) and the effects of fomulation and in vitro release conditions on the release of preparation.METHODS The rapid release core tablets of lansoprazole were prepared by wet granulation technique. The pH and enzyme double-sensitive LPZ-CT tablets were prepared by direct compression and sparying coating technique.RESULTS The rapid release core tablets dissolution in vitro was up to 70%.With 7%coating weight of pH layer and 20%guar gum of compressed layer, the drug in LPZ-CT tablet did not release in the simulated stomach and small intestine medium. In the simulated colon medium(pH7.4,0.2 g·L-1β-mannase),the cumulative release was more than 80%.CONCLUSION LPZ-CT tablets achieved the design requirements of in vitro drug release and precision targeting.
