Simultaneous estimation of lansoprazole and domperidone in combined pharmaceutical formulation using RHPLC
Abstract
A simple, precise and accurate high-performance liquid chromatographic (RHPLC) method was developed and validated for the simultaneous estimation of Lansoprazole (LSP) and Domperidone (DPD) in combined pharmaceutical dosage forms. Chromatographic separation was achieved using a C18 reversed-phase column with a mobile phase comprising acetonitrile and phosphate buffer (pH 7.0) in the ratio of 60:40 v/v. The flow rate was maintained at 1.0 mL/min and detection was carried out at 285 nm using a UV detector. The retention times of Lansoprazole and Domperidone were found to be approximately 3.4 min and 5.2 min, respectively. The method exhibited good linearity in the concentration range of 5–50 µg/mL for both drugs, with correlation coefficients (r²) greater than 0.999. Validation of the method was performed as per ICH guidelines, confirming the method’s accuracy, precision, robustness and specificity. The proposed method was successfully applied to the quantitative analysis of LSP and DPD in marketed tablet formulations.
