Bone Marrow Stromal Cells of Goat with Type Ι Osteoporosis Differentiate into Adipocytes and Osteoblasts
Abstract
The differentiation of type Ι osteoporosis goat bone marrow stromal cells into adipocytes and osteoblasts is investigated. We established The animal model of type Ι osteoporosis by performing ovariectomy on the 2-year-old female goat is establised. MSCs were isolated by all bone marrow culture method. The 2nd passage of MSCs were induced by adipogenic inducer and osteogenic inducer. The differentiation of adipocytes or osteoblasts were observed by oil red O, ALP and alizarin red staining. After 21 days of adipocytes inducing, the ratio of the BMSCs which displayed accumulation of lipid vacuoles were 81% After 14 days of osteogenic inducing, the rates of ALP positive cells were 78%. When cultured to 35 days, there were calcium node depositions. BMSCs of type Ι osteoporosis goat can be induced to adipocytes and osteoblasts.
