Effects of calcitonin on osteoblasts in vitro
Abstract
Objective To observe the effects of calcitonin(Miacalcic),an inhibitor of osteoclasts,on osteoblasts in vitro. Methods The culture medium with different Miacalcic concentrations(10 -4 ~10 -12 g/ml) and the second generation osteoblasts(OB-2) from the skull of newborn SD rats were mixed for the observation respectively on the proliferation(OD value at wavelength 570nm),the differentiation(ALP activity)and the mineralization(mineralized nodes/field of vision)of the OB-2. Results The OD values(mean and standard deviation)were 0^323±0^101~0^523±0^158;the ALP activity(mean and standard deviation) was 0^104±0^012 U/mg proteins, and the mineralized nodes(mean and standard deviation)were 5^75±0^957. Conclusion In comparison with control groups, the proliferation, differentiation and mineralization of the OB-2 were promoted by calcitonin concentration(10 -8 ~10+{12}g/ml),in which the effect of 10~10 g/ml calcitonin concentration on OB2 was the most significant for promoting proliferation( P 0 05),differentiation( P 0 01) and mineralization( P 0 001).
