Effects of emodin on osteoblast in vitro
Abstract
Aim To investigate the effects of emodin on the proliferation and differentiation of rat osteoblast in vitro. Methods To assess the effect of emodin on the osteoblasts, we isolated osteoblasts from rat calvaria. The osteocalcin secretion and mineralization of osteoblast were measured using radioimmunoassay and alizarin red stain respectively. Cell cycle was analyzed using flow cytometry and the mRNA expression of Type Ⅰ Collagen(COLL-I) in osteoblast were detected using reverse transcription-polymerase chain reaction(RT-PCR). Results Emodin enhanced supernatant osteocalcin and intracellular osteocalcin of osteoblast. Emodin up-regulated COLL-ⅠmRNA expression in osteoblast at concentrations of 2.5×10 -7 mol·L -1 and 5×10 -7 mol·L -1. However, emodin down-regulated COLL-ⅠmRNA expression in osteoblast at high concentrations of emodin. Emodin had no significant effect on cell cycle at concentrations from 2.5×10 -7 mol·L -1 to 2.5×10 -6 mol·L -1 and had no significant effect on mineralization at concentrations from 5×10 -7 mol·L -1 to 2.5×10 -6 mol·L -1. Conclusion Emodin can stimulate osteocalcin secretion and up-regulate COLL-ⅠmRNA expression of osteoblast in vitro.
