Abstract

By inhibiting sodium-glucose transporter-2 in the renal proximal tubule, new antidiabetic drugs known as gliflozins facilitate glucose and sodium elimination. This not only leads to a significant improvement in glucose control, but also to reductions in blood pressure and body weight in both diabetic patients and experimental models. Our study was designed to determine whether these beneficial effects could also be achieved in a model of L-NAME-induced non-diabetic hypertension, specifically in male Wistar rats. Over a period of six weeks, rats were administered empagliflozin at a dose of 10 mg/kg/day. Throughout the experiment, blood pressure was measured by tail cuff method, with one series measured by telemetry. In addition, renal parameters, body weight and amount of adipose tissue were analyzed. At the end of experiment, we investigated the role of the major vasoactive systems in the maintenance of blood pressure. Administration of empagliflozin showed a tendency to lower blood pressure. However, there was a minimal effect on body weight. Empagliflozin demostrated a positive effect on proteinuria and a slight decrease in plasma urea levels. Importanstly empagliflozin attenuated sympathetic component of blood pressure. Thus, we can conclude that empagliflozin produced favourable results in...
