Abstract

fluoro (6)-quinolone with potent antimicrobial activity against common respiratory pathogens (11).LVFX and GRNX show similar antimicrobial activities against gram-negative bacteria. However, GRNX has higher antimicrobial activity than LVFX against gram-positive bacteria, including staphylococci, streptococci, and pneumococci (12).Additionally, GRNX has higher broad spectrum antimicrobial activity against anaerobes than LVFX (13).These data suggest that GRNX may be an attractive agent for the treatment of CAP.However, few studies have investigated the antimicrobial activity of GRNX in mixed-infections containing aerobes and anaerobes. In the present study, we compared the antimicrobial activities of GRNX and LVFX in a murine model of mixed species bacterial pneumonia induced with S. pneumoniae and P. micra. MATERIALS AND METHODSAntimicrobial agents: Analytical grade powders of GRNX (FUJIFILM Toyama Chemical Co., Ltd., Tokyo, Japan) and LVFX (Sigma-Aldrich, Tokyo, Japan) were utilized throughout this study.GRNX and LVFX powders were suspended and diluted in 0.5% carboxymethyl cellulose (CMC-Na), to achieve desired concentrations, immediately prior to each experiment.Doses of GRNX and LVFX were determined from previous reports (14).Microorganisms: S. pneumoniae isolate (D-6888) was provided by the Scientific Research Network in Japan (SRNJ, Tokyo, Japan). The P. micra isolate (No.SUMMARY: Community-acquired pneumonia (CAP) can involve mixed-species bacterial infection. However, few…
