Abstract

Migraine is one of the most common neurological disorders which is usually associated with recurrent attacks of moderate to severe headaches along with nausea, photophobia, and phonophobia causing a considerable reduction in quality of life. The use of the conventional therapy, such as rizatriptan belongs to triptan family of drugs which shows some limitations like delay in action because of gastric stasis, variability in bioavailability and recurrence of symptoms. This paper is dedicated to the development of novel combination of dosage form of rizatriptan benzoate and caffeine with the aim of increasing the effectiveness by means of synergistic effect. The novel drug was created by applying the method of direct compression and then tested through the stages of preformulation, physicochemical investigations and evaluation of in vitro dissolution. As a result of the experiments, it can be concluded that the developed formulation showed good characteristics in terms of flowability, tablet features, as well as biphasic mode of action with quick initial effect followed by sustained one. No changes were observed during stability studies performed under accelerated conditions concerning both content and physical characteristics of the drug.
