Abstract

The present investigation focused on the formulation and evaluation of cubosome-loaded raft-forming liquid containing Monoammonium Glycyrrhizinate for gastro-retentive drug delivery. A Plackett–Burman design was employed to screen and evaluate the influence of formulation and process variables, including xanthan gum, gellan gum, glyceryl monooleate (GMO), poloxamer concentration, calcium carbonate concentration, sonication time, and stirring time on the critical quality attributes of the formulation. A total of 12 experimental formulations were prepared and characterized for floating duration, viscosity, percentage drug release, raft strength, particle size, and entrapment efficiency. The prepared formulations exhibited floating duration between 8–22 hours, viscosity ranging from 320.5–890.1 cP, drug release of 70.05–85.50% within 10 hours, raft strength between 1.4–3.8 g, particle size ranging from 140–470 nm, and entrapment efficiency between 50.590.5%. The results indicated that formulation variables significantly affected the physicochemical and gastro-retentive properties of the cubosomal raft system [1]. The developed raft-forming cubosomal formulations demonstrated desirable floating behavior, sustained drug release, suitable nanosized particle distribution, and good entrapment efficiency, suggesting their potential application in effective gastric retention and controlled delivery of Monoammonium Glycyrrhizinate for gastric ulcer management.
