Evaluation of the effect of metformin as a radiosensitiser in solid tumours: A systematic review
Abstract
Highlights • Metformin has demonstrated in preclinical studies its ability to enhance the effect of radiotherapy.• Metformin combined with radiotherapy, showed a significant benefit in survival and response to treatment in prostate, liver and gynecological tumors.• The benefict results in lung, rectal and head and neck cancer were inconsistent.• The mechanisms of action are improving the efficacy of radiotherapy by activating the AMPK pathway, inhibiting mTOR and reducing glutathione levels.• Despite promising results, the review highlights the lack of randomized clinical trials confirming the efficacy of metformin as a radiosensitizer in clinical practice.
How strong is the evidence?
Evidence Hub reads this as a systematic review. Its evidence level is 1 of 5 (Highest · pooled evidence). Pooled evidence from several studies sits at the top of the evidence pyramid.
