Combined Synthesis and Co-Amorphisation of Paracetamol with Citric Acid via Twin-Screw Extrusion (TSE)
Abstract
Due to the enhanced kinetic solubility and thus improved bioavailability of amorphous APIs, a reliable industrial amorphisation process is sought. Herein, we report, for the first time, the use of Twin Screw Extrusion (TSE) for the combined scalable synthesis of paracetamol and its subsequent co-amorphisation with citric acid, below their respective melting points and binary mixture eutectic temperature. The work illustrates the utility of TSE for combining synthesis and processing into a single operation.
