Abstract

This narrative review summarizes selected evidence on drug-resistant tuberculosis and the 6-month all-oral regimen of bedaquiline, pretomanid, linezolid, and moxifloxacin recommended by the World Health Organization. It covers global burden estimates, the TB-PRACTECAL randomized trial, an operational cohort in routine care in Uzbekistan and Belarus, and a trial of linezolid dosing. Unfavorable outcomes and serious adverse events were much less frequent with the short regimen than with standard care in the trial, and treatment success in routine care was high. The evidence is encouraging but limited in size and in comparative data, and success depends on rapid testing, drug supply, and toxicity monitoring. Some national figures come from news summaries and should be checked against WHO sources. Figures are quoted as reported; no data were pooled. This is a non-peer-reviewed preprint.
