Abstract

Introduction: Radiation-induced lung injury (RILI) is one of the most prevalent issues for patients receiving radiotherapy for lung and esophageal tumors. The present study intended to investigate the consolidative influence of onion oil on the protective effect of tadalafil drug (TDF) against the early phase of RILI in male rats. Methods: After 7 days of administration of onion oil (200 mg/Kg), TDF (6 mg /Kg), and their combination to different groups, the experimental animals were exposed to total body exposure to a single dose of gamma irradiation (6 Gy) to induce lung injury. Results: Data showed that irradiation caused a significant oxidative stress state in lung tissues, as revealed by an elevation in malondialdehyde (MDA) content, concomitant with a significant decline in antioxidant enzyme levels. Furthermore, it raised pro-inflammatory mediators, including tumor necrosis factor alpha (TNF-α), prostaglandin E2 (PGE2), cyclooxygenase-2 (COX-2), and intercellular adhesion molecule-1 (ICAM-1), accompanied by a reduction in the antiinflammatory cytokine interleukin 10 (IL-10). Western blotting analysis confirmed notable apoptosis progression in lung tissues, manifested by the elevation of Protein 53 (P53), Bcl-2- associated protein X (Bax), caspase-3 (Casp-3), B-cell lymphoma-2 (Bcl-2), and apoptosisinducing factor (AIF) apoptotic biomarkers. Discussion: Combined administration of (onion oil + TDF) provided more beneficial lung tissue protection, manifested by…
