External Cavity Quantum Cascade Laser Vibrational Circular Dichroism Spectroscopy for Fast and Sensitive Analysis of Proteins at Low Concentrations
Abstract
Vibrational circular dichroism (VCD) is a powerful mid-infrared spectroscopic technique that not only enables identification of enantiomers but is also highly sensitive to protein folding and protein secondary structure. While until recently VCD was limited by low intensity light sources which lead to long measurement times and low sensitivity, novel external cavity quantum cascade laser based VCD instrumentation significantly improves sensitivity and throughput. Enabling the recording of VCD spectra of the amide I and II spectral region of as little as 2 mg mL-1 of protein within 5 s.
